Studies of Some Novel Anti-Mycobacterium Agents:

Tuberculosis (TB) is a fatal infectious disease caused by bacterium call Mycobacterium tuberculosis. Each year the disease kills almost 2 million populations. The disease spreads due to unhygienic conditions also the bacterium develops resistance to the existing drugs (Multiple Drug Resistance-TB and Extensively Drug Resistance- TB). World Health Organization (WHO) has predicted by the Year 2020 there will be one billion new active cases if effective anti-TB drugs are not developed. The essential objectives of anti- tuberculosis therapy includes, fast killing of keenly multiply bacteria, avoidance of drug resistance and sterilization of infected host tissues to stop clinical relapse. In this study, we report few of the challenges such as drug resistance, duration of treatment, complexity of treatment in developing anti-tuberculosis drug along with their mode of action and limitations with reference to drug Isoniazid.  This project attempts to bring out the review of anti-TB drugs, and presents a novel method of synthesizing new anti-tuberculosis drugs and potential compounds to overcome the bacterial resistance and combat the re-emergence of tuberculosis. New anti-tuberculosis drugs  may show promising anti-tuberculosis activity.

Synthesis of some new Antituberculosis drugs:

The prolonged use of classical drugs developed a growing resistance and these drugs have gradually become less effective and incapable to meet the challenges, especially those of MDR-TB, XDR-TB, and HIV-TB co-infections.  Such MDR-TB is difficult and expensive to treat and is not always curable. Several nicotinic and isoniazid derivatives, most of them containing nitro groups were synthesized by our group, and evaluated for their in vitro antibacterial activity against Mycobacterium tuberculosis H37Rv using the Alamar Blue susceptibility test and the activity expressed as the minimum inhibitory concentration (MIC) in mg/mL. The novel method of synthesizing new anti-tuberculosis drugs(synthetic molecules) successfully obtained potential compounds for the treatment of tuberculosis. Therefore this class of compounds could be a good starting point to develop new lead compounds in the treatment of multi-drug resistant tuberculosis. It suggests that this class of compounds may be selectively targeted to M. tuberculosis growth, also considering that they were not cytotoxic to host cells at the same concentration and could be a good starting point to find new lead compounds.

Totally 11 new Anti-tuberculosis drugs were Synthesized and MIC of 25 mg/L was obtained. Snehal Bomble 2014 Batch BE Student of Chemical Engineering worked on project. She participated in Avishkar 2013, Chemcon 2013,  and Prakalp 2013.

Value added products from Karanja using novel techniques:

Karanja is abundantly found in most part of India. It had wide applications in pharmaceutical industries and considered as the key source for the synthesis of FAME. The aim of work is the extraction of value added products Karanjin from Karanja. It has application in the treatment of cancer. The intensified approach of ultrasound was found to be useful for enhancing the yield of Karanjin from the oil. The physical effects of ultrasound such as micro-emulsification, acoustic streaming reduced the diffusion limitations between the solid and the solvent phases.

Process Intensification of heterogeneously catalyzed for biodiesel synthesis for sustainable feedstock using a continuous hybrid reactor:

The objectives of the work were to develop the intensified strategies for esterification and transesterification processing for the synthesis of FAME from the sustainable feed stocks on a batch scale using cavitational and microwave reactor. The results of work confirmed that the intensified approach based on the microwave and sonochemical effect increased the rate of transesterification process on multi-fold basis compare to individual and conventional approaches. The continuous approach has been developed to enhance the rate of transesterification and esterification processing. The continuous approach eliminated the limitations of the low penetration depth of microwave and sonochemical effects and reduced the energy requirement for synthesis of FAME.

Biomass Fermentation in a 5 L Stirring Bioreactor:

 Fermentation typically is the conversion of carbohydrates to alcohols and carbon dioxide or organic acids using yeasts, bacteria, or a combination thereof, under aerobic or anaerobic conditions. Batch experiments were carried out on brown berries juice biomass. The crushed filtered juice of brown berries was sterilized by heating in a pressure cooker at around 1200C for half an hour. The glucose content of the juice was around 4%. The sterilized juice was then fermented in a conical flask at various temperatures and pH for 48 hours by adding sufficient nutrients and the developed culture medium containing Saccharomyces cerevisiae. Saccrification experiments were carried out which increased the glucose content by a minor amount of 1%. Final yield of ethanol obtained was around 6% as obtained at a pH of 5 and 350C.
